John Mulaney’s Champion Challenge (also knows as Boss Rush) has 2 chapters. The first chapter allows you to do each path separately, but the second one force you to complete all paths in one go. All paths have the same champions but their health and attack increase as you progress.

Top Counters to the Defenders

Howard the Duck

You need a champion that can nullify furies triggered by Rage Node. Also useful to nullify armors triggered by Explosive Personality. Here are the champions you can use

Doctor Doom
Symbiote Supreme
The Hood
Or any other champions that nullify buffs

Gwenpool

You need a Champion that either applies damage overtime debuff or gain power

Nick Fury
Hyperion
Vision Arkus
Sunspot
Venom
Guillotine 2099 (Can charge her to make Warlock easy)
Or any other that can apply debuff or gain power

Warlock

Tech Champions gain fury that increases attack by 125%. You better use a Tech champion, but not necessary. It’s not mandatory but better to remove willpower and regeneration master to fight Warlock, else you will degenerate lots of health and your power will also be drained many times.

Or any champ that you like. Can be done with almost any champ.

Deadpool

Just use a champion that can stop healing. Or any hard-hitter also works.

I used CMM for the first four paths but she failed to solo on the last 2 because of the high health pool.

You need a champion that is immune to Poison and can heal block (Masochism Healing). The best team to use

Captain America Infinity War (200 Sigs) with Skill and Tech Synergy

Spider-Ham – The Boss

You need a champion that can stop evade.

When Spider-Ham Evades, you get a fatigue debuff on you. While fatigue debuff is applied, you suffer 25% of the defender’s attack each time you land a hit. Your 10% power is drained every second while it is applied. Additionally, each time your power empties to Zeo, suffer +240% of the defender’s attack as direct damage.

Vision Aarkus (Just parry and wait for Cold Snap to apply)
Nick Fury
She-Hulk
Falcon
Or any champ that causes 100% ability accuracy or stops evade
